TLR4/NF- κBp65 the Effect of High Expression of on the Proliferation, Invasion, and Metastasis of Skin Squamous Cell Carcinoma

Abstract: Objective To explore and analysis the effects of Toll like receptor 4 (TLR4)/nuclear factor NF-κBp65 high expression on the proliferation, invasion, and metastasis of skin squamous cell carcinoma. Methods Human skin squamous cell carcinoma A431 cell line with logarithmic growth phase were divided into three groups - blank group, control group, and experimental group. They were transfected with DMEM medium, pc3.0 empty vector, and pc3.0-NF-κBp65, respectively vector. All the cells were used to detect the expression of proliferation, invasion, and metastasis in skin squamous cell carcinoma. Results After 24 and 48 hours of transfection, the expression of TLR4 and NF-κBp65 in the experimental group were significantly higher than that of the blank group and control group (P<0.05), and there were no significant difference compared between the blank group and control group (P>0.05). The cell proliferation index of the experimental group were significantly lower than that of the blank group and control group (P<0.05), and there were no significant difference compared between the control group and the blank group (P>0.05). The cell invasion index and cell metastasis index of the experimental group were significantly lower than those of the blank group and control group (P<0.05), while there were no significant difference compared between the control group and the blank group (P>0.05). The expression level of Caspase-9/Myc protein in the experimental group were significantly higher that of the blank group and control group (P<0.05), and there were no significant difference compared between the blank group and control group (P>0.05). Conclusion TLR4/NF- κBp65 high expression can inhibit the proliferation, metastasis, and invasion of skin squamous cell carcinoma cells, as well as promote the expression of Caspase-9/Myc protein, thereby exerting promote apoptotic effect on tumor cells.
