Extracting Steroidal Saponins of Chinese Chive Seeds and Anti-fatigue Activities Atudying

Abstract: Objective To identify the steroidal saponins from Chinese chive seeds as raw material by ethanol, and to determine the extraction optimization conditions to obtain the best extraction conditions and determine the anti-fatigue activity. Methods Chinese chive seeds were extracted by 50% ethanol, separated by MCI column, and determined whether the 70% ethanol eluate contained steroidal saponins by color development reactions: Liebermann-Burchard (LB) colorimetric assay, Kahlenberg colorimetric assay, Rosen-Heimer colorimetric assay, and the effect of steroidal saponins of Chinese chive seeds on the exhaustive swimming time and blood lactic acidin(LAC) was studied by Yang deficiency mouse model. Results The 70% ethanol eluate of Chinese chive seeds contained steroidal saponins, and the mean value of total saponin content of Chinese chive seeds was 3.28% under 50% ethanol, 60℃, and 1:25 material to liquid ratio for 4 hours, which was the best extraction condition and could significantly prolong exhaustive swimming time(P<0.05)and reduce blood lactic acid content in Yang-deficient mice (P<0.05). Conclusion Chinese chive seed steroidal saponin extract can improve the state of exercise fatigue in mice and has significant anti-fatigue activity.
