Sanzi Yangqin Decoction Attenuates Lung Injury in Asthmatic Mice by Regulating JAK/STAT Signaling Pathway

Abstract: Objective To study the effects of Sanzi Yangqin Decoction attenuating lung injury in asthmatic mice by regulating JAK/STAT signaling pathway. Methods Fifty mice were randomly divided into normal group, model group, positive group, low-dose Sanzi Yangqin Decoction group and Sanzi Yangqin Decoction high-dose group. Ovalbumin was used to replicate the asthma mouse model. HE staining was used to detect mouse lung tissue, and to detect lung function.IL-6 and TNF-α levels in BALF were measured by ELISA,The protein expression levels of JAK, p-JAK, STAT3 and p-STAT3 were detected by Western Blotting. Results The normal group of mice has neat organization structure, clear alveolar structure, thin alveolar walls, no inflammatory cell infiltration in the alveoli, and rare collagen deposition in the alveolar epithelium and interstitium. Inflammatory cell infiltration, there is a lot of collagen deposition in the alveolar interstitium. The pathological improvement of the lung tissue in the positive group and the low-dose and high-dose groups of Sanzi Yangqin Decoction was improved compared with the model group, and the pathological improvement of the lung tissue in the high-dose Sanzi Yangqing Decoction group was the most obvious. Compared with the normal group, the FEV1 and PEF of the mice in the model group were significantly decreased (P<0.05); compared with the model group, the FEV1 and PEF of the positive group, the low-dose Sanziyangqin Decoction group and the high-dose Sanziyangqin Decoction group were significantly lower (P<0.05). were significantly increased (P<0.05). Compared with the normal group, the levels of IL-6, TNF-α, p-JAK, and p-STAT3 in the model group were significantly increased (P<0.05). The levels of IL-6, TNF-α, p-JAK and p-STAT3 in the dose group and the high-dose Sanziyangqin decoction group were significantly decreased (P<0.05). Conclusions Sanzi Yangqin Decoction can effectively improve the related pathological changes in asthmatic mouse models, and its mechanism of action may be to improve the inflammatory response of the lungs by regulating the JAK/STAT signaling pathway.
