Effect of Up Regulating miR-590-3p on Invasion and Metastasis of Pancreatic Cancer Cells and Its Mechanism

Abstract: Objective To observe the effect of miR-590-3p gene expression on the invasion and metastasis of human pancreatic cancer Capan-2 cells. Methods Human pancreatic cancer Capan-2 cells were divided into two groups: negative control group (NC group) and miR-590-3p up-regulated group (miR-590-3p mimics group). The expression of miR-590-3p in human normal pancreatic ductal epithelial cells hTERT-HPNE and pancreatic cancer Capan-2 cells was detected by qRT PCR. The effect of miR-590-3p on the apoptosis of Capan-2 cells was detected by flow cytometry, the effect of miR-590-3p on the proliferation of Capan-2 cells was detected by MTT test, the change of cell migration ability was detected by scratch test, and the change of cell invasion ability was detected by Transwell test, the expression of AKT/mTOR pathway related proteins was detected by Western blot. Results The expression level of miR-590-3p in Capan-2 cells of pancreatic cancer was significantly lower than that in normal pancreatic duct epithelial cells, and the expression level of miR-590-3p mRNA in miR-590-3p mimics group was significantly higher than that in NC group (P<0.05). The results of flow cytometry showed that the level of apoptosis in miR-590-3p mimics group was significantly higher than that in NC group (P<0.05). The results of cell scratch and invasion test showed that the migration and invasion ability of miR-590-3p mimics group was significantly lower than that of NC group (P<0.05). Western blot results showed that compared with the NC group, the levels of p-AKT and p-mTOR proteins in the miR-590-3p mimics group were significantly reduced. Conclusion Up regulation of miR-590-3p expression can increase the apoptosis of pancreatic cancer cells and inhibit the migration and invasion of pancreatic cancer cells.
